ABSTRACT: The impact of estradiol on endothelial dysfunction: An experimental model in hypercholesterolemic rats. Summary there is a higher risk of cardiovascular diseases in males than in females. This has generated growing scientific interest in the role of sex hormones in the different stages of atherogenesis. Endothelial dysfunction is an early process present from the onset of atherosclerosis and accompanies its evolution and complications, with the key involvement of sex hormones. Estrogen therapy increases the bioactivity of nitric oxide (NO), a protector of the endothelium, in the vascular system of healthy postmenopausal women. However, the role of estrogens in the endothelial dysfunction observed in endothelium-independent relaxation induced by nitroglycerin is not completely clarified. This study aims to determine whether the deterioration of vascular function, particularly the endothelial dysfunction observed during endothelium-independent vasodilation, can be reversed through the administration of estrogens (E2) in the aortic tissue of rats. The findings could optimize and develop new therapies to prevent and treat cardiovascular diseases. The study demonstrated that estradiol decreases the deterioration of endothelium-independent relaxation in the aorta of hypercholesterolemic rats due to the reduction of calcium ion activity in smooth muscle cells. In an experimental model with 27 Sprague Dawley albino rats, the results revealed that the administration of estradiol significantly prevents endothelial dysfunction and improves the vasodilatory response.
